Fura-2, pentasodium salt is a cell-impermeant, fluorescent ion indicator for intracellular calcium ion measurements. Fura-2’s spectral response to ion-binding permits a ratiometric calculation of ion concentration that cancels artifacts of photobleaching, dye concentration gradients, cell thickness and dye leakage. The pentasodium salt form of Fura-2 is water soluble and may be loaded into live cells by microinjection, patch pipette infusion, scrape loading, bead loading, or ATP-induced permeabilization. The excitation wavelength of Fura-2 varies with Ca²⁺ concentration and shifts to shorter wavelengths with increasing Ca²⁺ concentration. The ratio of fluorescence intensity at 510 nm when excited at 340/380 nm is used for ratiometric calculation of Ca²⁺ concentration. This is the pentasodium salt form of Fura-2 for use in applications that may be perturbed by potassium ions. .
