SBFI, tetraammonium salt is a cell-impermeant, fluorescent ion indicator that is relatively selective for sodium ions in the presence of other mono-valent cations. SBFI’s spectral response to ion-binding permits a ratiometric calculation of ion concentration that cancels artifacts of photobleaching, dye concentration gradients, and dye leakage. SBFI has been used to investigate intracellular sodium concentrations and gradients in a variety of living tissues. SBFI, tetraammonium is water soluble and may be loaded into live cells by microinjection, patch pipette infusion, scrape loading, bead loading, or ATP-induced permeabilization. The excitation wavelength of SBFI varies with sodium ion concentration and shifts to shorter wavelengths with increasing sodium concentration. The ratio of fluorescence intensity at 500 nm when excited at 340/380 nm, as well as the fluorescence emission ratio at 410/590 nm with excitation at 340 nm have both been used for ratiometric calculation of ion concentration. .
