6-TAMRA is a purified single isomer, amine-reactive orange-fluorescent dye for synthesis of orange-fluorescent biomolecules. This single isomer dye has been widely used in preparing fluorescent nucleotides and oligonucleotides and is one of the classical fluorophores used in automated DNA sequencing. 6-TAMRA carboxylic acid may be reacted with amines on peptides, proteins, modified nucleotides, biopolymers and other amine-containing biomolecules using a carbodiimide like EDAC or DIPC to yield a robust amide linkage. Carboxylic acids may also be used to form ester-linked conjugates or acid-halide products. The absorbance spectrum of some 6-TAMRA conjugates may be dependent on the degree of labeling density and dye-dye interactions, but the fluorescence is insensitive to environment pH and is relatively photostable. Conjugates of 6-TAMRA are excited near 540 nm with fluorescence emission near 568 nm.
