DiI stain is a lipophilic yellow-fluorescent dye that is commonly used as a retrograde neuronal tracer dye and as a long-term label for cell lineage tracing. DiI stain is also commonly used as a membrane probe for investigations of membrane fusion and cell membrane dynamics. DiI has an environmentally dependent fluorescence that is greatly enhanced when the dye is incorporated into membranes or bound to lipophilic biomolecules while it is weakly fluorescent in aqueous environments. DiI is excited near 550 nm and has an environmentally dependent fluorescence emission near 565 nm.
