Indo-1, pentapotassium salt is a cell-impermeant fluorescent ion indicator for intracellular calcium ion measurements in live cells. Indo-1’s fluorescence response to ion-binding permits a ratiometric calculation of calcium ion concentration that cancels artifacts of photobleaching, dye concentration gradients, cell thickness and dye leakage. When excited at 350 nm, Indo-1’s fluorescence emission wavelength shifts from near 475 nm to near 400 nm upon binding of calcium ions. Indo-1 has been used as a fluorescent calcium ion indicator in a wide variety of living cells and tissues and is well-suited for use in flow cytometry applications. The pentapotassium salt form of Indo-1 is water soluble and may be loaded into live cells by microinjection, patch pipette infusion, scrape loading, bead loading, or ATP-induced permeabilization. .
