Mag-fura-2, tetrapotassium salt is a cell-impermeant, fluorescent ion indicator for intracellular magnesium ion measurements. Mag-fura-2’s spectral response to ion-binding permits a ratiometric calculation of ion concentration that cancels artifacts of photobleaching, dye concentration gradients, cell thickness and dye leakage. The tetrapotassium salt form of Mag-Fura-2 is water soluble and may be loaded into live cells by microinjection, patch pipette infusion, scrape loading, bead loading, or ATP-induced permeabilization. The excitation wavelength of Mag-fura-2 varies with magnesium ion concentration and shifts to shorter wavelengths with increasing magnesium concentration. The ratio of fluorescence intensity at 510 nm when excited at 340/380 nm is used for ratiometric calculation of ion concentration. .
