EMA is an orange-fluorescent, light-activated photoaffinity label for nucleic acids that does not readily cross the intact membranes of most live cells. EMA may be used a cell viability probe that can enter cells with compromised plasma membranes where it then binds to nucleic acids as an intercalator. The nucleic acid bound EMA can then be photolyzed by light to yield a covalent linkage between the dye and the nucleic acids. DNA that has been covalently labeled with EMA will not be amplified in PCR techniques, providing a method to suppress amplification of extracellular DNA and DNA from dead cells (viability-PCR, or v-PCR). EMA also exhibits a fluorescence that is greatly enhanced when bound to nucleic acids and EMA stained cells may be fixed using aldehyde-based methods. Nucleic acid bound EMA is excited near 504 nm and has an environmentally dependent fluorescence emission near 600 nm. .
