Fluo-3 AM is a green-fluorescent ion-indicator that is selective for Ca2+ ions. Fluo-3 responds to Ca2+ binding with an increase in fluorescence intensity. Live cells are passively loaded with Fluo-3 AM by incubation with the cell membrane-permeant AM esters. Once internalized, cytosolic esterases cleave the AM esters to liberate the active Fluo-3 tetra-carboxylate ion indicator. After enzymatic hydrolysis, Fluo-3 is optimally excited near 506 nm with fluorescence emission near 526 nm. The longer wavelength of absorption is convenient for minimizing photodamage in cells. Fluo-3 AM is useful for fluorescence microscopy, confocal microscopy, flow cytometry, and HCS applications. Fluo-3, AM has proved particularly valuable for determining GPCR function via cell-based high-throughput screening. Fluo-3 AM has relatively low water solubility and Pluronic F-127 (#6361), a mild detergent, is often used as a dispersing agent to facilitate the loading. .
