DiOC5(3) is a green-fluorescent, lipophilic dye for staining cell membranes that exhibits a slow fluorescence response to membrane potential in live cells. The fluorescence of this dye is environmentally dependent and is greatly enhanced when incorporated into membranes or bound to lipophilic biomolecules. Hyperpolarization of cell membrane potential results in decreasing fluorescence emission due to formation of dye aggregates with reduced fluorescence. DiOC5(3) is excited near 483 nm and has an environmentally sensitive emission near 500 nm.
