Dibromobimane is a low molecular weight, non-ionic, fluorescent probe for labeling thiols and crosslinking proteins. This cell-permeant probe is essentially non-fluorescent until both bromomethyl groups have reacted with thiols. The bromomethyl groups are relatively selective for reaction with thiols at physiological pH but may also react with carboxylates at pH above 7. Dibromobimane has been used to form an intramolecular-crosslink in myosin, hemoglobin, ATPase and other thiol-containing proteins. The conjugates of Dibromobimane are optimally excited near 394 nm and have a variable emission near 490 nm.
