NDA is a fluorogenic derivatization agent that permits ultra-sensitive detection limits for primary amines. NDA is non-fluorescent until it reacts with a primary amine in the presence of an excess of a nucleophile like cyanide or a thiol like mercaptoethanol. NDA forms stable adducts with amines on biomolecules like peptides and proteins and has been used extensively for derivatization of amines for analysis by HPLC and capillary electrophoresis. NDA may be used as a pre-column or post-column derivatization and NDA derivatives can be excited near 420 nm or by a He-Cd laser line near 442 nm.
