5(6)-TAMRA is an amine-reactive orange-fluorescent dye for synthesis of orange-fluorescent biomolecules. This dye has been widely used in preparing fluorescent peptides, proteins, nucleotides, oligonucleotides, and enzymes. 5(6)-TAMRA carboxylic acid may be reacted with amines on peptides, proteins, modified nucleotides, biopolymers and other amine-containing biomolecules using a carbodiimide like EDAC or DIPC to yield a robust amide linkage. Carboxylic acids may also be used to form ester-linked conjugates or acid-halide products. The absorbance spectrum of some TAMRA conjugates may be dependent on the degree of labeling density and dye-dye interactions, but the fluorescence is insensitive to environment pH and is relatively photostable. Conjugates of 5(6)-TAMRA are excited near 540 nm with fluorescence emission near 572 nm.
