Calcein Blue, AM is a cell-permeant, blue-fluorescent, cell viability probe and polar tracer dye for short-term staining of live cells. Live cells are passively loaded with weakly fluorescent Calcein Blue, AM by incubation with the cell membrane-permeant AM esters. Once internalized, cytosolic esterases cleave the AM esters to liberate the dicarboxylate, polar tracer dye. Calcein Blue’s fluorescence is enhanced after enzymatic hydrolysis and the excitation and emission wavelengths shift to slightly longer wavelengths, 360 / 449 nm respectively. Caclein Blue AM is intrinsically fluorescent and Calcein Blue stained cells may require an extra wash step to reduce background fluorescence. .
